Abstract
EVALUATION OF FREE RADICAL SCAVENGING ACTIVITY OF CRATAEGUS MACRACANTHA LODD FLOWERS (Abstract): The antioxidant activity of biological products has been demonstrated by various “in vitro” and “in vivo” studies. Aim: To measure the capacity of an extract obtained from Crataegus macracantha Lodd flowers to capture free radicals from chlorpromazine (radical indicators), in order to evaluate the entire antioxidant activity. Material and methods: The lyophilisate from Crataegus macracantha Lodd flowers were obtained in the Department of Pharmacognosy of the Iaşi “Grigore T. Popa” University of Medicine and Pharmacy. The radical indicators were obtained through oxidation of chlorpromazine with potassium persulphate in acid environment. The diminution of radical solution color was determined by means of spectrophotometry (maximum absorbance at ʎ=525 nm), according to the amount of lyophilisate. Ascorbic acid was used as a standard antioxidant. Results: The antioxidant activity was evaluated by using the calibration curve, obtained by plotting the radical solution absorbance variation according to ascorbic acid concentration. Due to the fact that antioxidant activity is simultaneously influenced both by the antioxidant concentration and response time, a variant of the method using the calibration curve obtained by plotting the area under the curve (AUC), against ascorbic acid concentration. AUC was calculated for each ascorbic acid concentration by graphic representation of the absorbance inhibition percentage at every two predefined time frames. The antioxidant activity of the sample is calculated by using the regression line equation (r2=0.999) and was expressed in micromolar equivalents ascorbic acid/g product (59,70 µM/g). Conclusions: The obtained results confirm the capacity to capture free radicals and, implicitly, the antioxidant activity of Crataegus macracantha Lodd flowers.
