EVALUATION OF NEUROLOGIC EFFECTS OF SOME NEW IBUPROFEN THIAZOLIDIN-4-ONE DERIVATIVES

Abstract

Nonsteroidal anti-inflammatory drugs (NSAIDs), such as ibuprofen, are widely used to treat pain and inflammation associated with various conditions, including neurodegenerative diseases. The aim of this study was to evaluate the neurological effects of several new ibuprofen derivatives containing a thiazolidin-4-one moiety. Materials and methods. The effects of ibuprofen-thiazolidine-4-one derivatives (4a-n) on neuromotor function and locomotor activity in mice were assessed using the Rota-rod and locomotor activity tests. The compounds were administered orally by gavage, as a suspension in tween 80, at a single daily dose equivalent to 1/20 of the LD50. A reference group, which received ibuprofen, and a control group, which received tween 80, were also used. Results: The ibuprofen-thiazolidine-4-one derivatives (4a-n) did not produce statistically significant changes in neuromotor performance, at any of the three time points assessed (1 hour, 2 hours, 6 hours post-administration). However, a significant decrease in total locomotor activity-both horizontal and vertical movements—was observed with compound 4b. Notably, several derivatives also led to a marked reduction in vertical movements. Conclusions: The observed decrease in vertical movements for some ibuprofen thiazolidine-4-one derivatives may indicate a potential anxiolytic effect, though this hypothesis requires validation through further studies.
